The Mitochondrial Protocol: SS-31, MOTS-C & Epitalon
Precisely sequenced three-compound protocol targeting cellular energy, metabolic signalling, and longevity pathways.
Why Mitochondria?
Mitochondria are not simply power generators — they are signalling organelles that communicate with the nucleus, regulate inflammation, govern cell survival, and modulate metabolic rate systemically. Mitochondrial output declines with age: the electron transport chain becomes less efficient, ROS production increases, NAD+ levels fall, and cardiolipin degrades. This protocol addresses three different aspects of that decline through three compounds with distinct but complementary mechanisms.
Storage: All three compounds must be refrigerated after reconstitution. Room temperature storage after reconstitution degrades the compounds.
Reconstitution Reference
| Compound | Vial Size | BAC Water | Dose Conversion |
|---|---|---|---|
| SS-31 (Elamipretide) | 10mg | 1ml | 10iu = 1mg |
| MOTS-C | 40mg | 2ml | 10iu = 2mg |
| Epitalon | 50mg | 2ml | 20iu = 5mg |
Phase 1 — SS-31: Mitochondrial Foundation (Days 1-20)
SS-31 (Elamipretide) selectively concentrates in the inner mitochondrial membrane by binding to cardiolipin — a phospholipid unique to mitochondria. This allows it to quench ROS exactly at their production site, protecting electron transport chain complex integrity and improving ATP synthesis efficiency. Running SS-31 alone for 20 days builds the mitochondrial baseline before MOTS-C and Epitalon are layered in.
| Compound | Dose | Timing |
|---|---|---|
| SS-31 (Elamipretide) | 1mg (10iu) | Daily — morning. Refrigerate after reconstitution. |
Phase 2 — MOTS-C: Metabolic Activation (Day 21+)
MOTS-C is encoded within mitochondrial DNA itself — making it a true retrograde mitochondrial signal. It activates AMPK (the master cellular energy sensor), shifting metabolism toward fat oxidation, improving insulin sensitivity, and enhancing exercise capacity. Pre-workout timing maximises the metabolic demand intersection. From Day 41 dosing shifts to every other day as adaptation occurs.
| Period | Dose | Frequency |
|---|---|---|
| Days 21-40 | 2mg (10iu) | Daily — pre-workout |
| Day 41+ | 2mg (10iu) | Every other day |
Phase 3 — Epitalon: The Longevity Layer (Days 31-50)
Epitalon is a synthetic tetrapeptide originally developed by Professor Vladimir Khavinson. Its primary mechanism is telomerase activation — the enzyme that maintains telomere length, which shortens with each cell division and is closely linked to cellular aging. It also regulates the pineal gland's melatonin production, relevant for circadian rhythm and sleep architecture. The protocol is exactly 20 days with a minimum 4-month rest — this is not a precaution, it is the documented protocol design.
| Compound | Dose | Timing | Duration |
|---|---|---|---|
| Epitalon | 5mg (20iu) | Daily — before bed | 20 days total. 2 × 50mg vials. Then 4+ months off. |
Full Protocol Timeline
Do not extend Epitalon beyond 20 days. The 4-month rest is part of the protocol design, not a precaution. Running it continuously or longer is not supported by existing research literature.
